摘要 | We present a novel droplet-based barcoded combinatorial Screening (BaCon) platform which enables rapid, massively parallel, and ultra-high-throughput screening of optimal conditions of cell-free protein expression systems. With this platform, we first demonstrate the ability of BaCon in recognizing microdroplets under various reaction conditions using fluorescent labeling. We further apply this technique to optimize a cell-free protein synthesis system. Broadly, BaCon can recognize multiparameter mixtures possessing any optically assayable function or morphological change, including optimization of reagent and reaction conditions, combinatorial drug screening, directed evolution, and biochemical assays. © 2022 MicroTAS 2022 - 26th International Conference on Miniaturized Systems for Chemistry and Life Sciences.
