摘要 | We report logarithmic-dilution-based droplet digital PCR (LD3 PCR), a customized yet generalizable droplet digital PCR (ddPCR) approach that features a significantly extended dynamic range over 7 orders of magnitude. Such a performance is 3 orders higher than conventional ddPCR and close to well-established quantitative PCR (qPCR). LD3 PCR was used for HPV16 genotyping in clinical-relevant samples. The results show that LD3 PCR maintains accuracy and robustness in detecting 40 to 1.98×107 copies/μl templates. We anticipate that LD3 PCR may be broadly applied in quantifying clinical-relevant viral pathogens. © 2022 MicroTAS 2022 - 26th International Conference on Miniaturized Systems for Chemistry and Life Sciences.
